Program

student in a laboratorium

The online course DNA Sequencing - NGS & TGS will provide an overview of the sequencing technologies Illumina, Ion Torrent, Nanopore & PacBio. The goal is to understand library preparation, PCR amplification and short- and long-read sequencing. You will also learn how sequencing data is processed, visualized and interpreted. Combining theory and practice will help you develop an understanding of the technology and how it is applied in data analysis.

Learning outcomes

  • You will have insight into the principles of Next Generation Sequencing (NGS) and Third Generation Sequencing (TGS);
  • You will understand the advantages and limitations of both sequencing technologies;
  • You will know how raw sequencing data are processed;
  • You will know how the results are visualized;
  • You will know how to interpret sequencing data.

Topics

The course covers the following topics:

Day 1 (theory)
  • Principes of DNA sequencing
  • Next-generation sequencing (NGS):
    - Technologies: Illumina & Ion Torrent
    - Library preparation
    - PCR amplification
    - Short read sequencing
  • Third generation sequencing (TGS):
    - Technologies: Oxford Nanopore & PacBio
    - Long read sequencing
  • Comparison of NGS & TGS: advantages and limitations
Day 2 (theory)
  • Data processing: from raw data to usable information
  • Data quality and data preprocessing
  • Read mapping versus genome assembly
  • Applications of NGS & TGS
Day 3 (practice)
  • Deep dive into NGS & TGS data
  • Quality control of NGS & TGS data
  • Read mapping of NGS data
  • Visualization of read mapping data
  • Bring your own data

Participants’ own data
On the third day of the course you will be given data to work with. You can also bring your own data, provided they meet the requirements that the lecturer provides in class.
